RESUMO

The vigorous development of marine fisheries carbon sinks (MFCS) has become a momentous pathway to mitigate global warming and effectively cope with the climate crisis. Deservedly, based on clarifying mechanism of carbon sequestration, this paper designs a research paradigm for predicting and evaluating the potential of MFCS. Specifically, a novel nonlinear grey Bernoulli model, namely MFCSNGBM(1,1), is proposed by innovatively mining the original data law through adaptive cumulative series and introducing the compound Simpson formula to optimize background values. More precisely, we utilize a heuristic Grey Wolf Optimization algorithm to find the best power index, which enhances the adaptability. To prove usefulness and robustness of MFCSNGBM(1,1) model, yields of seven common shellfishes (oyster, clam, mussel, scallop, razor clam, bloody clam, and snail) and three main algae (kelp, pinnatifid undaria, and laver) are predicted and compared with six competing models. Based on prediction results, new model has the most accurate predictions, with all prediction errors being <10 %, and thus can achieve effective prediction of shellfish and algae production from 2022 to 2025. Further, the capacity and potential of MFCS in China are scientifically evaluated using a removable carbon sink model, considering various yield levels and biological parameters of shellfish and algae. The assessment results show that during the sample period, China's marine fisheries carbon sinks steadily increased with an annual growth rate of 57,000 tons. From 2022 to 2025, with support of policy of MFCS and improvement of disaster prevention and mitigation capacity, the potential of MFCS will be further released. The growth rate of MFCS will be increased to 94,000 tons per year, and its overall scale is expected to reach 2,198,245 tons by 2025, equivalent to fixing 8.06 million tons of CO2. The carbon sink's economic value is significantly estimated to be over 400 billion yuan.

BACKGROUND: In the United States, patients with limited English proficiency face significant barriers to comprehending and acting upon health-related information, particularly during the COVID-19 pandemic. The ability of health professionals to communicate COVID-19-related information to Mandarin-speaking patients has proved critical in discussions about vaccine efficacy, side effects, and post-vaccine protection. METHODS: The authors created a one-hour educational module to help Mandarin-speaking medical students better convey COVID-19 vaccine information to Mandarin-only speakers. The module is composed of an educational guide, which introduced key terminology and addressed commonly asked questions, and pre- and post-surveys. The authors recruited 59 Mandarin-speaking medical students all of whom had previously completed a medical Mandarin elective. The module and surveys were distributed and completed in August 2021. Data analysis measured the change in aggregate mean for subjective five-point Likert-scale questions and change in percent accuracy for objective knowledge-based questions. RESULTS: 86.4% of participants were primary English speakers with variable levels of Mandarin proficiency. The educational module significantly improved participants' subjective comfort level in discussing the COVID-19 vaccine in English and Mandarin. The largest improvement in both English and Mandarin was demonstrated in participants' ability to explain differences between the COVID-19 vaccines, with an aggregate mean improvement of 0.39 for English and 1.48 for Mandarin. Survey respondents also demonstrated increased percent accuracy in knowledge-based objective questions in Mandarin. CONCLUSIONS: This module provides Mandarin-learning medical students with skills to deliver reliable information to the general population and acts as a model for the continued development of educational modules for multilingual medical professionals.

According to the survey statistics, most traffic accidents are caused by the driver's behavior and status irregularities. Because there is no multi-level dangerous state grading system at home and abroad, this paper proposes a complex state grading system for real-time detection and dynamic tracking of the driver's state. The system uses OpenMV as the acquisition camera combined with the cradle head tracking system to collect the driver's current driving image in real-time dynamically, combines the YOLOX algorithm with the OpenPose algorithm to judge the driver's dangerous driving behavior by detecting unsafe objects in the cab and the driver's posture, and combines the improved Retinaface face detection algorithm with the Dlib feature-point algorithm to discriminate the fatigue driving state of the driver. The experimental results show that the accuracy of the three driver danger levels (R1, R2, and R3) obtained by the proposed system reaches 95.8%, 94.5%, and 96.3%, respectively. The experimental results of this system have a specific practical significance in driver-distracted driving warnings.

Topology optimization holds great potential to achieve the best performance for various fluid-related applications like aircraft components and microfluidic mixers. This paper reviews comprehensively the technical progress of this field over the last decade from the viewpoint of structural expression. The density-based approach has been widely adopted to design structures due to its simple concept, ease of implementation, and robustness. Different designs using such a pointwise method for systems under Stokes, laminar Navier-Stokes, turbulent, non-Newtonian, and steady-state/unsteady-state fluid flows are reviewed and discussed in depth. Both isogeometric analysis and the moving morphable components/voids methods will demonstrate their advantages regarding integration with computer-aided design. The moving morphable components/voids method also significantly reduces computing costs. From the viewpoint of boundary smoothness, we are most concerned about whether smoother boundaries can reduce objective functions such as energy dissipation even lower. Therefore, this work also concentrates on level set and spline expression methods. Furthermore, we identify isogeometric analysis and machine learning in shaping the field's future. In addition, the review highlights the following two challenges: achieving accurate fluid model construction and the relatively limited experimental validation of fluid topology optimization outcomes.

Electrocardiogram (ECG)-gated multi-phase computed tomography angiography (MP-CTA) is frequently used for diagnosis of coronary artery disease. Radiation dose may become a potential concern as the scan needs to cover a wide range of cardiac phases during a heart cycle. A common method to reduce radiation is to limit the full-dose acquisition to a predefined range of phases while reducing the radiation dose for the rest. Our goal in this study is to develop a spatiotemporal deep learning method to enhance the quality of low-dose CTA images at phases acquired at reduced radiation dose. Recently, we demonstrated that a deep learning method, Cycle-Consistent generative adversarial networks (CycleGAN), could effectively denoise low-dose CT images through spatial image translation without labeled image pairs in both low-dose and full-dose image domains. As CycleGAN does not utilize the temporal information in its denoising mechanism, we propose to use RecycleGAN, which could translate a series of images ordered in time from the low-dose domain to the full-dose domain through an additional recurrent network. To evaluate RecycleGAN, we use the XCAT phantom program, a highly realistic simulation tool based on real patient data, to generate MP-CTA image sequences for 18 patients (14 for training, 2 for validation and 2 for test). Our simulation results show that RecycleGAN can achieve better denoising performance than CycleGAN based on both visual inspection and quantitative metrics. We further demonstrate the superior denoising performance of RecycleGAN using clinical MP-CTA images from 50 patients.

OBJECTIVE: Conventional cervical lymph node dissection often leaves large surgical scars, which seriously compromises the postoperative aesthetic effect and can affect the quality of life of patients. In this study, the safety and feasibility of robotic-assisted endoscopic thyroidectomy with central neck dissection (CND) and lateral neck dissection (LND) via a combined transoral and breast approach are discussed in detail. MATERIALS AND METHODS: A retrospective analysis was made of the data of 26 patients with stage cN1b papillary thyroid carcinoma who were admitted to the Thyroid Surgery Department of the Hunan Cancer Hospital from March 2021 to September 2022 and who underwent robotic-assisted endoscopic thyroidectomy with LND via a combined transoral and breast approach. The demographic data, surgical indicators, postoperative data, and the postoperative complication rate of the patients were analyzed, and the learning curve was analyzed by cumulative summation. RESULTS: All the patients underwent endoscopic surgery without any conversion to open surgery. The mean operation time was 313.7±50.3 min and the mean number of total positive/retrieved lymph nodes was 11.2±8.1/36.8±13.7. Two patients developed temporary laryngeal recurrent nerve palsy and three patients developed temporary hypoparathyroidism, all of whom recovered within 3 months postoperatively. No tumor recurrence occurred during follow-ups that ranged from 6 to 24 months. The mean postoperative quality of life (QOL) score was 189.1±118.2, test results ranging from 0 to 1300 with a lower score indicating a higher QOL, and the aesthetic satisfaction score was 4.2±0.7, test scores ranging from 0 to 5 with higher scores indicate higher satisfaction. The turning point of the learning curve was in the 11th case. CONCLUSIONS: The robotic-assisted endoscopic thyroidectomy with CND and LND via a combined transoral and breast approach is safe and feasible, and the improved cosmetic effect is remarkable, which is conducive to improving the postoperative QOL of patients. It provides a new surgical option for patients.

BACKGROUND: Rewriting the genomes of living organisms has been a long-standing aim in the biological sciences. The revelation of the CRISPR/Cas9 technology has revolutionized the entire biological field. Since its emergence, this technology has been widely applied to induce gene knockouts, insertions, deletions, and base substitutions. However, the classical version of this system was imperfect for inducing or correcting desired mutations. A subsequent development generated more advanced classes, including cytosine and adenine base editors, which can be used to achieve single nucleotide substitutions. Nevertheless, these advanced systems still suffer from several limitations, such as the inability to edit loci without a suitable PAM sequence and to induce base transversions. On the other hand, the recently emerged prime editors (PEs) can achieve all possible single nucleotide substitutions as well as targeted insertions and deletions, which show promising potential to alter and correct the genomes of various organisms. Of note, the application of PE to edit livestock genomes has not been reported yet. RESULTS: In this study, using PE, we successfully generated sheep with two agriculturally significant mutations, including the fecundity-related FecBB p.Q249R and the tail length-related TBXT p.G112W. Additionally, we applied PE to generate porcine blastocysts with a biomedically relevant point mutation (KCNJ5 p.G151R) as a porcine model of human primary aldosteronism. CONCLUSIONS: Our study demonstrates the potential of the PE system to edit the genomes of large animals for the induction of economically desired mutations and for modeling human diseases. Although prime-edited sheep and porcine blastocysts could be generated, the editing frequencies are still unsatisfactory, highlighting the need for optimizations in the PE system for efficient generation of large animals with customized traits.

The care delivered to patients with cardiovascular disease involves coordination among a multitude of clinical team members spanning diverse inpatient and outpatient settings. The majority of quality improvement interventions in cardiovascular care have been developed based on quantitative evidence, which neither fully accounts for multilevel determinants (eg, patient, clinician, and institution) nor contextualization from key informants. The rigor and effectiveness of these interventions would be enhanced by mixed-methods studies whose strengths include (1) the use of qualitative research methodologies (eg, eliciting patient or clinician perspectives on barriers and facilitators of best practices) and (2) integrating qualitative and quantitative data and analyses to understand more fully effective strategies for achieving optimal care and outcomes for these patients across diverse settings. This article illustrates the application of a complex mixed-methods design to advance an evidence-based, customizable infection prevention toolkit for durable left ventricular assist device therapy. The study (1) uses quantitative clinical data merged with Medicare claims to evaluate interhospital variability in the incidence of infection; (2) uses qualitative methods to understand local practice patterns across low- and high-performing centers; and (3) integrates both data sources to gain a comprehensive understanding of the overall findings.

BACKGROUND: Left ventricular assist device (LVAD) implantation leads to substantial and sustained improvement in health-related quality of life (HRQOL) among patients. Infection following device implantation remains an important and frequent complication and adversely affects patient-reported HRQOL. METHODS: Patients in The Society of Thoracic Surgeons' Interagency Registry for Mechanically Assisted Circulatory Support receiving a primary LVAD between April 2012 to October 2016 were included. The primary exposure was one-year post-implant infection, characterized by: (1) any infection; (2) total number of infections and (3) type (LVAD-specific, LVAD-related, non-LVAD). The association between infection and the primary composite adverse outcome (defined as EuroQoL Visual Analog Scale< 65, too sick to complete the survey, or death at 1-year) was estimated using inverse probability weighting and Cox regression. RESULTS: The study cohort included 11,618 patients from 161 medical centers with 4,768 (41.0%) patients developing an infection, and 2,282 (19.6%) patients having> 1 infection during the follow up period. The adjusted odds ratio for the primary composite adverse outcome was 1.22 (95% CI, 1.19-1.24, p < 0.001) for each additional infection. Each additional infection was associated with a 3.49% greater probability of the primary composite outcome and was associated with worse performance across multiple dimensions of HRQOL as assessed by the EQ-5D for patients who survived to 1 year. CONCLUSIONS: For patients undergoing LVAD implantation, each additional infection within the first post-implantation year was associated with an incremental negative effect on survival free of impaired HRQOL.

Apple scar skin viroid (ASSVd) can infect apple trees and cause scar skin symptoms. However, the associated physiological mechanisms are unclear in young saplings. In this study, ASSVd-infected and control 'Odysso' and 'Tonami' apple saplings were examined to clarify the effects of ASSVd on apple tree growth and physiological characteristics as well as the leaf metabolome. The results indicated that leaf ASSVd contents increased significantly after grafting and remained high in the second year. Leaf size, tree height, stem diameter, branch length, and leaf photosynthetic efficiency decreased significantly in viroid-infected saplings. In response to the ASSVd infection, the chlorophyll a and b contents decreased significantly in 'Odysso', but were unchanged in 'Tonami'. Moreover, the N, P, K, Fe, Mn, and Ca contents decreased significantly in the leaves of viroid-infected 'Odysso' or 'Tonami'. Similarly, the CAT and POD contents decreased significantly in the viroid-infected saplings, but the SOD content increased in the viroid-infected 'Tonami' saplings. A total of 15 and 40 differentially abundant metabolites were respectively identified in the metabolome analyses of 'Odysso' and 'Tonami' leaves. Specifically, in the viroid-infected 'Odysso' and 'Tonami' samples, the L-2-aminobutyric acid, 6″-O-malonyldaidzin, and D-xylose contents increased, while the coumarin content decreased. These metabolites are related to the biosynthesis of isoflavonoids and phenylpropanoids as well as the metabolism of carbohydrates and amino acids. These results imply that ASSVd affects apple sapling growth by affecting physiological characteristics and metabolism of apple leaves. The study data may be useful for future investigations on the physiological mechanisms underlying apple tree responses to ASSVd.

How to increase meat production is one of the main questions in animal breeding. Selection for improved body weight has been made and, due to recent genomic advances, naturally occurring variants that are responsible for controlling economically relevant phenotypes have been revealed. The myostatin (MSTN) gene, a superstar gene in animal breeding, was discovered as a negative controller of muscle mass. In some livestock species, natural mutations in the MSTN gene could generate the agriculturally desirable double-muscling phenotype. However, some other livestock species or breeds lack these desirable variants. Genetic modification, particularly gene editing, offers an unprecedented opportunity to induce or mimic naturally occurring mutations in livestock genomes. To date, various MSTN-edited livestock species have been generated using different gene modification tools. These MSTN gene-edited models have higher growth rates and increased muscle mass, suggesting the high potential of utilizing MSTN gene editing in animal breeding. Additionally, post-editing investigations in most livestock species support the favorable influence of targeting the MSTN gene on meat quantity and quality. In this Review, we provide a collective discussion on targeting the MSTN gene in livestock to further encourage its utilization opportunities. It is expected that, shortly, MSTN gene-edited livestock will be commercialized, and MSTN-edited meat will be on the tables of ordinary customers.

OBJECTIVE: Endothelial function may improve with enhanced external counterpulsation (EECP) or acupuncture. This study aimed to evaluate the feasibility of acupoint stimulation combined with EECP (acupoint-EECP) for endothelial cell function in patients with essential hypertension. METHODS: Thirty essential hypertensive patients were, randomly divided into two groups, with 15 patients in the acupoint-EECP group, and 15 patients in the control group, of which 3 cases were lost by week 6. Both groups were treated with continued medicine. The participants in the acupoint-EECP group received acupoint stimulation combined with EECP therapy, 45 min for each time, 5 times weekly for 6 weeks for a total of 22.5 hours. The selected acupoints are Zusanli (ST36), Fenglong (ST40) and Sanyinjiao (SP6). The curative effects of the two groups were compared. RESULTS: The acupoint-EECP group (n=15) showed significant improvement in endothelial function [nitric oxide (NO) ,endothelin-1 (ET-1) and carotid-femoral pulse wave velocity (cf-PWV), respectively] values compared to the control group (n=12). Multiple imputation (n = 20 imputations) was performed to account for potential bias due to missing data. In stratified analyses, SBP and DBP values decreased when the baseline SBP was ≥120 mmHg and DBP was ≥80 mmHg. CONCLUSIONS: These findings suggest the feasibility of acupoint-EECP in improving endothelial function and treating hypertension. Floral fragrance is an important trait that contributes to the ornamental properties and pollination of crabapple. However, research on the physiological and molecular biology of the floral volatile compounds of crabapple is rarely reported. In this study, metabolomic and transcriptomic analyses of the floral volatile compounds of standard Malus robusta flowers (Mr), and progeny with strongly and weakly fragrant flowers (SF and WF, respectively), were conducted. Fifty-six floral volatile compounds were detected in the plant materials, mainly comprising phenylpropane/benzene ring-type compounds, fatty acid derivatives, and terpene compounds. The volatile contents were significantly increased before the early flowering stage (ES), and the contents of SF flowers were twice those of WF and Mr flowers. Odor activity values were determined for known fragrant volatiles and 10-11 key fragrant volatiles were identified at the ES. The predominant fragrant volatiles were methyl benzoate, linalool, leaf acetate, and methyl anthranilate. In the petals, stamens, pistil, and calyx of SF flowers, 26 volatiles were detected at the ES, among which phenylpropane/benzene ring-type compounds were the main components accounting for more than 75% of the total volatile content. Functional analysis of transcriptome data revealed that the phenylpropanoid biosynthesis pathway was significantly enriched in SF flowers. By conducting combined analyses between volatiles and differentially expressed genes, transcripts of six floral scent-related genes were identified and were associated with the contents of the key fragrant volatiles, and other 23 genes were potentially correlated with the key volatile compounds. The results reveal possible mechanisms for the emission of strong fragrance by SF flowers, and provide a foundation for improvement of the floral fragrance and development of new crabapple cultivars.

Polyploid plants are usually salt tolerant, but the underlying mechanisms remain fragmental. This study aimed to dissect salt resistance of tetraploid honeysuckle (Lonicera japonica Thunb.) from ion balance, osmotic adjustment and antioxidant defense by contrasting with its autodiploid through pot experiments. Less salt-induced reduction in leaf and root biomass confirmed higher tolerance in tetraploid honeysuckle, and moreover, its greater stability of photosynthetic apparatus was verified by mild influence on delayed chlorophyll fluorescence transients. Compared with the diploid, greater root Na+ exclusion helped alleviate salt-induced decrease in leaf K+/Na+ for maintaining ion balance in tetraploid honeysuckle, and relied on Na+/H+ antiporter activity, because their difference of root Na+ exclusion disappeared after applying a specific inhibitor of Na+/H+ antiporter. Lower reduction in leaf relative water content suggested higher tolerance to osmotic pressure in tetraploid honeysuckle under salt stress, which hardly resulted from osmotic adjustment given the similar decrease extent of leaf osmotic potential with that in the diploid. In contrast to significant elevated leaf lipid peroxidation and superoxide dismutase and ascorbate peroxidase activities in the diploid, no obvious changes in them suggested that tetraploid honeysuckle never suffered salt-induced oxidative stress. According to more accumulated leaf chlorogenic acid and phenolics and greater elevated leaf phenylalanine ammonia-lyase activity and transcription, leaf phenolic synthesis was enhanced greater in tetraploid honeysuckle upon salt stress, which might serve to prevent oxidative threat by consuming reducing power. In conclusion, polyploidy enhanced salt tolerance in honeysuckle by maintaining ion homeostasis and water balance and preventing oxidative stress.
